Cell-type-specific role of P2Y2 receptor in HDM-driven model of allergic airway inflammation

Allergic airway inflammation (AAI) is a chronic respiratory disease that considered severe restriction in daily life and accompanied by constant risk of acute aggravation. It characterized IgE-dependent activation mast cells, infiltration eosinophils, activated T-helper cell type 2 (Th2) lymphocytes into mucosa. Purinergic receptor signaling known to play crucial role inducing maintaining allergic inflammation. Previous studies an ovalbumin (OVA)–alum mouse model demonstrated contribution the P2Y2 purinergic subtype (P2RY2) However, conflicting data concerning mechanism which P2RY2 triggers AAI has been reported. Thus, we aimed at elucidating cell-type-specific house dust mite (HDM)-driven Thereupon, HDM-driven was induced conditional knockout mice, deficient or intact for P2ry2 either alveolar epithelial hematopoietic myeloid helper T dendritic cells. To analyze functional these mice models, flow cytometry bronchoalveolar lavage fluid (BALF), cytokine measurement BALF, invasive lung function measurement, HDM re-stimulation mediastinal lymph node (MLN) histology were performed. Mice subjected HDM-based resulted reduced signs including eosinophilia peribronchial inflammation, Th2 production, bronchial hyperresponsiveness Furthermore, migration bone-marrow-derived cells monocytes, both , towards ATP impaired. Additionally, found levels MCP-1/CCL2 IL-8 homologues BALF lower concentrations IL-33 tissue In summary, our results show contributes HDM-induced mediating proinflammatory production drives recruitment monocytes.
